چکیده
ABSTRACT Membrane contact sites (MCSs) between the endoplasmic reticulum (ER) and late endosomes/lysosomes (LE/lys) are emerging as critical hubs for diverse cellular events, changes in their extents linked to severe neurological diseases. While recent studies show that synaptotagmin-like mitochondrial-lipid-binding (SMP) domain-containing protein PDZD8 may mediate formation of ER–LE/lys MCSs, functions remain largely elusive. Here, we attempt investigate lipid transfer activities extent which its depend on activities. In accordance with studies, demonstrate is a protrudin (ZFYVE27)-interacting acts tether at MCSs. Furthermore, discover SMP domain binds glycerophospholipids ceramides both vivo vitro, can transport lipids membranes vitro. Functionally, required LE/lys positioning neurite outgrowth, dependent activity domain. This article has an associated First Person interview first author paper.

Dystrophin Dp71 is required for neurite outgrowth in PC12 cells.
To determine the role of Dp71 in neuronal cells, we generated PC12 cell lines in which Dp71 protein levels were controlled by stable transfection with either antisense or sense constructs. Cells expressing the antisense Dp71 RNA (antisense-Dp71 cells) contained reduced amounts of the two endogenous Dp71 isoforms. Antisense-Dp71 cells exhibited a marked suppression of neurite outgrowth upon the ...
متن کاملA grab to move on: ER-endosome contacts in membrane protrusion formation and neurite outgrowth.
A key feature of many eukaryotic cells, most prominently seen in developing neurons, is their ability to form and extend membrane protrusions. How protrusion formation is linked to exocytic membrane trafficking is largely unclear. In a recent paper published in Nature, Raiborg et al identify a crucial role in this process for dynamic membrane contact sites (MCSs) between the ER and endosomes. T...
متن کاملThe tumor suppressor protein p53 is required for neurite outgrowth and axon regeneration.
Axon regeneration is substantially regulated by gene expression and cytoskeleton remodeling. Here we show that the tumor suppressor protein p53 is required for neurite outgrowth in cultured cells including primary neurons as well as for axonal regeneration in mice. These effects are mediated by two newly identified p53 transcriptional targets, the actin-binding protein Coronin 1b and the GTPase...
متن کاملThe BTB domain of the nuclear matrix protein NRP/B is required for neurite outgrowth.
The neuronal nuclear matrix protein, NRP/B, contains a BTB domain and kelch repeats and is expressed in primary neurons but not in primary glial cells. To examine the function of NRP/B in neurons, we analyzed the structure/function of the NRP/B-BTB domain and its role in neurite outgrowth. Based on three-dimensional modeling of NRP/B, we generated an NRP/B-BTB mutant containing three mutations ...
